Revisiting Preadolescent Cardiomyocyte Proliferation in Mice.

چکیده
Understanding cardiomyocyte cell cycle regulation after birth is key to optimizing regenerative strategies for the heart post injury, yet poses multiple technical challenges, as evidenced by recent studies that have arrived at divergent conclusions. In a recent publication in Cell, Alkass et al undertook multiple approaches to examine cardiomyocyte cell cycle regulation in the first 3 weeks after birth. Here, we summarize results of Alkass et al and 3 other groups in examining preadolescent cardiomyocyte cell cycle regulation, highlighting the distinct approaches and incumbent caveats.

No Evidence for Cardiomyocyte Number Expansion in Preadolescent Mice
The magnitude of cardiomyocyte generation in the adult heart has been heavily debated. A recent report suggests that during mouse preadolescence, cardiomyocyte proliferation leads to a 40% increase in the number of cardiomyocytes. Such an expansion would change our understanding of heart growth and have far-reaching implications for cardiac regeneration. Here, using design-based stereology, we ...

Cardiomyocyte Cell-Cycle Activity during Preadolescence
Earlier studies (Soonpaa et al., 1996) revealed a rapid drop-off of ventricular cardiomyocyte cell-cycle activity at birth in mice, followed by a burst of DNA synthesis during the first week of postnatal life which contributed to the formation of multi-nucleated cardiomyocytes by postnatal day 10 (PN10). It has recently been suggested that a second burst of cardiomyocyte cell-cycle activity occ...
متن کاملComparison of random and aligned PCL nanofibrous electrospun scaffolds on cardiomyocyte differentiation of human adipose-derived stem cells
Objective(s):Cardiomyocytes have small potentials for renovation and proliferation in adult life. The most challenging goal in the field of cardiovascular tissue engineering is the creation of an engineered heart muscle. Tissue engineering with a combination of stem cells and nanofibrous scaffolds has attracted interest with regard to Cardiomyocyte creation applications. Human adipose-derived s...
متن کاملA microRNA-Hippo pathway that promotes cardiomyocyte proliferation and cardiac regeneration in mice.
In contrast to lower vertebrates, the mammalian heart has limited capacity to regenerate after injury in part due to ineffective reactivation of cardiomyocyte proliferation. We show that the microRNA cluster miR302-367 is important for cardiomyocyte proliferation during development and is sufficient to induce cardiomyocyte proliferation in the adult and promote cardiac regeneration. In mice, lo...
